摘  要:目的评价冻融过程对胚胎透明带的影响。方法采用蛋白酶消化法对卵裂期胚胎进行消化,对酶消化前后胚胎的透明带厚度进行比较。结果在酶消化前,实验组和对照组胚胎的透明带厚度分别为(19·07±0·84)μm和(19·03±0·75)μm,差异无显著性(P>0·05)。在酶消化后,实验组和对照组胚胎透明带厚度分别为(13·01±0.88)μm和(8·70±0·73)μm。与消化前相比,两组胚胎透明带厚度均变薄,差异有显著性(P<0·001)。酶消化后,实验组与对照组相比,胚胎透明带厚度明显较厚,差异有显著性(P<0·001)。结论酶消化法可使新鲜胚胎和冻融胚胎透明带明显变薄。冻融胚胎较新鲜胚胎透明带难以被蛋白酶消化。Objective To evaluate the effect of freezing - thawing on the zona pellucida (ZP) of embryos. Methods A controlled experimental study design was employed. The embryos were classified into two groups according to their stages. 30 frozen-thawed embryos were used in the experiment group and 30 fresh embryos were used in the control group. All the embryos were not worth of undergoing embryo transfer. The ZP thickness of the embryos before and after ZP ablation with pronase was compared in the two groups. Results The ZP thickness of frozen - thawed embryos and fresh embryos before ablation with enzyme were ( 19.07 ± 0.84) μm and ( 19.03 ± 0. 75 ) μm in the experiment and control groups, respectively ( P 〉 0. 05 ) , and difference was not statistically significant. The ZP thickness of embryos after ablation with enzyme were ( 13.01 ±0. 88) μm and (8.70 ±0. 73) μm in the experiment and control groups, respectively. Compared to that of embryos before ablation with enzyme, the ZP thickness of the embryos in the two groups were significantly thinner (P 〈 0. 001 ). But the ZP thickness of the embryos in the experiment group was thicker than that of the control group after ablation with enzyme (P 〈 0. 001 ). Conclusion The ZP thickness of frozen-thawed embryos and fresh embryos becomes thinner after ablation with enzyme. The ZP of frozen-thawed embryos is more resistant to enzyme ablation compared to that of fresh embryos.
