摘 要:目的分析国内99株布鲁氏菌的遗传多态性,为其分型和诊断奠定基础。方法提取布鲁氏菌DNA,经内切酶Eco RI酶切、琼脂糖凝胶电泳、Southern转印后,与地高辛标记的16 srRNA核糖体基因探针杂交,以酶免疫试验检测信号,比较布鲁氏菌各标准株和野生株的16 srRNA核糖体指纹图谱。结果不同布鲁氏菌菌株在16 s rRNA探针杂交图谱上有明显的多态性,根据杂交的条带数及片段的大小差异,99株布鲁氏菌分属17个RT型(ribotype,RT型)。以RT15型为优势型,其他型别则较分散,有的RT型只存在于某个菌株。结论我国布鲁氏菌在遗传分化及克隆群的分布上存在多态性,提示国内布鲁氏菌在流行上具有复杂性。Objective To investigate the genetic polymorphism of 99 Bmcella strains isolated in China, so as to provide the basis for ribotyping and diagnosis of Brucella infection. Methods Chromosomal DNA of Brncella was digested with Eco RI, and separated by agarose gel electrophoresis and Southern blotting. Then it was tramsferred into a nylon filter and hybridized with 16 s rDNA which was labeled with digoxigenin-dUTP. Hybrid was detected by enzyme immunoassay. Compare the fingerprint of 16 s rRNA in different Brucella strains. Results 16 s rRNA fingerprint of different Brucella strains showed high polymorphism. 99 Brucella strains detected belonged to 17 ribotypes(RT) according to the numbers and locations of strips on 16 s rRNA fingerprint. RT 15, as the most predominant ribotype, comprised 26.3% Bmeella strains. The location of other ribotypes was scattered, some ribotype was only found in certain strain. Conclusions Brucella strains in China showed genetic polymprphism and clonal diversity, suggesting that the epidemic of Brucella is complicated.
