摘 要:目的:探讨当归对高浓度葡萄糖所致血管内皮细胞损伤保护作用的机制。方法:以不同浓度的葡萄糖(葡萄糖组,7.5-60 mmol/L)、不同浓度当归(当归组,12.5-200 mg/L)及联合组,当归联合24 h半数致死量(24hIC50)葡萄糖,30mmol/L作用于人体脐静脉内皮细胞(VEC-304)24 h,观察细胞生长状态、MTT法检测细胞增长率、RT-PCR检测细胞内皮生长因子(VEGF)mRNA表达及Western blot检测VEGF蛋白水平。结果:与对照组比较,不同浓度作用VEC-304 24h,葡萄糖组A值明显降低;当归组则明显升高(P<0.05或P<0.01);联合组于低浓度当归(12.5-100 mg/L)联合葡萄糖对细胞起抑制作用,高浓度(100-200 mg/L)当归联合葡萄糖则有增殖作用。RT-PCR和West-ern blot显示,葡萄糖组VEGF mRNA和蛋白表达下调;联合组则表达正常。结论:高浓度葡萄糖可损伤血管内皮细胞,当归可保护高浓度葡萄糖下血管内皮细胞,这可能是当归治疗糖尿病并发组织缺血性疾病的机制之一。Objective: To investigate the mechanism of protective effects of Angelica on vascular endothelial cell (VEC- 304) injury induced by high dose of glucose. Methods: Morphological changes of vascular endothelial cells (VEC-304) were observed in 3 groups after 24 h in vitro: various doses of glucose treated group (7. 5- 60 mmol/L), Angelica-treated group (12.5-200 mg/L), combined treatment group (glucose 30 retool/L, Angelica 12.5-200 mg/L). The growth rate was measured by MTT assay. The expression of VEGF mRNA and protein in VEC-304 was detected by RT-PCR and Western blot respectively. Results: After the VEC-304 was treated with different concentrations of glucose for 24 h, compared with control group, A values were significantly reduced, and when the VEC-304 was treated with different concentrations of Angelica for 24 h, A values were significantly increased (P〈0.05 or P〈0. 01). In the combined treatment group, low dose of angelica (12.5-100 mg/L) in combination with glucose could inhibit the growth of the cells, while high dose of Angelica (100- 200 mg/L) in combination with glucose could promote the proliferation of the cells. RT-PCR and Western blot revealed that the expression of VEGF mRNA and protein was down-regulated in glucose-treated group, but it was normal in combined treatment group. Conclusion: The growth of vascular endothelial cells could be inhibited by high concentration of glucose, while Angelica could protect the vascular endothelial cells exposure to high dose of glucose, which might he one of the mechanisms of Angelica treating diabetes associated with tissue ischemia diseases.
