摘  要:目的了解冻存复苏过程对保持软骨细胞生物学特性的影响。方法对冻存1周组、1个月组和新鲜制备组的软骨细胞进行倒置相差显微镜动态观察、生长曲线、HE染色以及Ⅱ型胶原免疫组化等观察,在其形态和功能方面进行比较。结果各组间软骨细胞的生长过程、Ⅱ型胶原免疫组化染色结果差异均无显著性,但随冻存时间的延长,其存活率有所下降。结论冻存软骨细胞与新鲜制备的软骨细胞相比保持了一些相似的生物行为,但冻存时间长短对冻存的软骨细胞存活率有影响。Objective To study the frozen effection to mandibular condylar cartilage cells. Methods Human mandibular condylar cartilage (MCC) cells were harvested and amplified by microcarrier culturetechnique MCC, cells were preserve in liquid nitrogen , The biological properties of cryopreserved MCC cells were investigated. The deferences of the type Ⅱ collagen were tested by SABC immunohistochemical of the type Ⅱ collagen after freezing; the mRNA levels were tested and so on the mRNA levels were tested by in situ hybridization of the type Ⅱ collagen between the transfection group and comparison group . Results Cells from liquid nitrogen preservation kept their phenotypic , proliferative an metabolic characteristics of MCC cells in vivo, The expression of collagen Ⅱ is stronger than the comparison group on the fifth generation. Conclusion freezing reservation of MCC cells can hold the same biological chacterical, but the life capacity of the frozen cells of rabbit mandibular condylar cartilage is lower than comparison group .
