摘　　要：目的对日本血吸虫Sj23DNA进行缺失突变,构建Sj23DNA突变体疫苗,为寻求更佳免疫效果的疫苗奠定基础。方法利用重叠PCR技术将Sj23DNA上与ME491同源的部分碱基缺失,获得Sj23DNA的突变体,将其克隆入真核表达载体pcDNA3的多克隆位点上,构建重组质粒pcD-NA3-Sj23突变体,进行酶切和测序鉴定,并用CLUSTALX和primerpremier软件将测序的结果与Sj23DNA序列进行分析。结果序列分析显示pcDNA3-Sj23突变体缺失序列与原设计一致,双酶切pcDNA3-Sj23突变体获得预期大小的DNA片段。结论成功构建了缺失与ME491同源部分碱基的质粒pcDNA3-Sj23突变体。Objective To find a more effective vaccine against schistosomiasis by constructing the deletion mutant of Sj23DNA. Methods A mutant of Sj23DNA without the homologous sequence of ME491 was obtained by overlap PCR and then it was inserted into eukaryotie expression vector pcDNA3 to construct the recombinant plasmid. The recombinant plasmid was examined by restriction enzyme digestion and sequence analysis. The sequence of the mutant of Sj23DNA was compared with Sj23DNA by using CLUSTAL X and the primer premier. Results The deletion sequence in the mutant of Sj23 DNA was identified to the former design by sequence comparison and restriction enzyme digestion. Conclusion The mutant of peDNA3-Sj23 without the homologous sequence of ME491 is constructed successfully.
