低渗培养对LLC-PK1细胞pICln蛋白分布的影响

摘　　要：目的探讨低渗条件下LLC-PK1细胞pIC ln蛋白的分布变化。方法分别用等渗和50%低渗培养基对LLC-PK1细胞进行培养,用抗pIC ln血清抗体对细胞可溶性和不溶性成分的pIC ln进行W estern b lot检测,同时进行细胞的荧光抗体染色。结果W estern b lot显示等渗培养时LLC-PK1细胞可溶性成分中pIC ln免疫反应带的密度为(51±3)%;50%低渗培养时明显增多到(69±3)%,两者比较有统计学意义(P<0.01)。荧光抗体染色也显示了pIC ln在胞质中明显增多。结论pIC ln对低渗是敏感的,它可能作为胞质调节蛋白、低渗时在胞质中分布增多能促进胞内渗透性物质的运出,参与调节细胞体积而对抗低渗。Objective To study the change of distribution of pICln in LLC-PKI ceils under low osmotic pressure. Methods LLC- PK1 cells were cultured with osmotic and 50% bypotonic medium respectively. The distribution of pICln were analyzed with anti-pICln serum in soluble and insoluble fraction by Western blot, at the same time, cells were stained by the method of immunofluorescence. Results The results of Western blot showed that the density of pICln bands was （51 ± 3 ） % and （69 ±3） % in soluble traction at osmotic and 50% hypotonic medium respectively（P 〈 0.01 ）. The stained pICln was much more numnerous in cytoplasmic fraction at the 50% hypotonic condition than that at the osmotic one.Conclusions PiClnis is sensitive to hypotonic condition, the increase of pICln in cytoplasmic fraction as cytoplasmic regulatory protein may reinforce the efflux of intracellular osmolyte, therefore regnlate the cell volume to resist hypotonic condition.
