摘  要:目的:研究小檗碱和环孢素联用时大鼠肝组织中肿瘤及免疫相关基因表达谱的影响。方法:大鼠予以盐酸小檗碱ig 6 d,最后一天予以环孢素单次ig给药。取肝脏提取总RNA。用Cy3和Cy5两种荧光物质分别标记阴性时照组,环孢素组和环孢素+盐酸小檗碱组肝脏mRNA,制备成探针与表达谱芯片进行杂交。结果:在被检测的4 096个基因中,环孢素组与肿瘤及免疫相关的差异表达基因共3个,均为表达下调。环孢素+盐酸小檗碱组与肿瘤及免疫相关的盖异表达基因共14个,其中表达上调的相关基因5个,表达下调的相关基因9个。结论:小檗碱和环孢素联用可影响大鼠肝组织中肿瘤及免疫相关基因的表达。Objective: To study the effects of coadministration of berberine(Ber) with cyclosporine(CsA) on tumor associated and immune associated genes expression in rat liver. Method: After orally treated with berberine for 6 days and orally treated with CsA in the last day, the rats were sacrificed and the liver was taken out to extract total RNA. mRNA from negative control group and CsA group and Ber +CsA group were labeled with Cy3-dCTP and CyS-dCTP separately. These probes were hybridized with cDNA microarray. Result: Among 4096 genes, 3 tumor associated and immune associated genes were down-regulated in CsA group and 14 tumor associated and immune associated genes were differentially expressed with 5 genes up-regulated and 9 genes down-regulated in Ber + CsA group, Conclusion: Coadministration of berberine (Ber) with eyclosporine (CsA) can effect the expression of the tumor associated and immune associated genes in rat liver.
