摘  要:对影响甘蓝型油菜与紫罗兰下胚轴原生质体融合后的融合体培养因素进行了研究。结果表明:采用液体浅层培养,以改良的KM8p为培养基附加2,4-D1.5 mg/L,NAA0.5 mg/L,6-BA0.5 ml/L,以蔗糖0.3 mol/L和葡萄糖0.1 mol/L作渗透稳定剂进行培养效果最好;原生质体培养3~6 d,细胞发生第1次分裂,第7天时分裂频率最高为13.8%;35天后形成大量的细胞团和肉眼可见的小愈伤组织,其植板率最高为6.5%。The breeding of crops depends on a lot of germplasm resources. Many hybrid plants were generated from inter-specific and intergeneric crossing. It is a new technology to expand germplasm resource of crops. In this paper the culture factors of fusion-protoplasts between Brassica nopus L. and Matthiola incana B were analyzed. The appropriate condition of fusion-protoplasts culture were that: the fusion-protoplasts were cultured in improved KM8p medium supplemented with 1.5 mg/L 2,4-D, 0.5 mg/L NAA and 0.5 ml/L BA with the method of "Thin Liquid Layer". 0.3 mol/L sucrose in combination with 0.1 mol/L glucose that was used as osmotic regulator were found superior to other combinations. The fusion-protoplast started to be divided after 3-6 days, and the division frequency was 13.8 % after being cultured for 7 days. The cell clone and small tallus could be obtained in 35 days and the plating efficiency was 6.5 %.
