摘 要:目的探讨聚乙二醇(PEG)法在生物带瓣管道准备中的应用价值,比较PEG去细胞前后带瓣管道的物理特性。方法猪主动脉瓣带瓣管道分为去细胞组和正常对照组,去细胞组用PEG和DNaseⅠ处理,苏木精-伊红染色观察去细胞情况,DNA含量测定,计算去细胞百分率;猪主动脉带瓣管道条置于力学测试仪,测定最大负荷、最大应力、最大应变和弹性模量。结果PEG组去细胞百分率为95.32%,去细胞猪带瓣管道组织中看不到细胞成分;且细胞外基质(ECM)结构保存完整,胶原纤维排列整齐,无明显断裂,仍呈波浪状平行排列,结构紧凑,弹性纤维结构清晰,组织无明显水肿。生物力学检测,与对照组比较最大负荷、最大应力、弹性模量、最大应变等的差异均无显著性意义。结论PEG法去除细胞完全,细胞外基质保存完整,对组织机械性能无明显影响,适于构建组织工程带瓣管道。Objective To study a new method of preparing acellular porcine valved conduits by decellularization with poly ethylene glycol and to observe the change of mechanical properties. Methods The porcine aortic conduits were decellularized with poly ethylene glycol (PEG) and DNase I , while the fresh porcine aortic conduits were used as control. Histological examination of them stained with HE dye was done. The content of DNA was detected in the two groups by photometer. The mechanical properties were assessed by AGS-J. Results The decellularrzing percentage was 95.32% and the structure of extra cellular matrix (ECM) was well reserved in PEG group. The mechanical properties had no significant difference between the two groups in peak load, maximal stress, etc. Conclusion The method of PEG could remove cellular components effectively, while it did not disrupt the histological structure of ECM and the mechanical properties.
