摘  要:从柔嫩艾美耳球虫长春株(由单卵囊接种建立)挑选出单孢子囊,制备成单孢子囊胶囊,给鸡接种后建立5个克隆,传代后提取DNA,用已筛选的13条随机引物进行PCR扩增。结果表明:每个克隆均获得RAPD图谱,有特异条带。5个克隆中C2和C3相似值为0.993 1,C4和C5相似值为0.992 7,C1与C2和C4的相似值分别为0.931 6和0.875 3。鉴别结果表明C1为1个克隆,C2和C3为1个克隆,C4和C5为1个克隆,说明柔嫩艾美耳球虫卵囊中的4个孢子囊的基因组成不一致。Five different clones, established by a single sporocyst infection from Changchun strain of Eimeria tenella, were studied by Random Amplified Polymorphic DNA(RAPD) with 13 screened random primers. The result showed that the RAPD map and the especial fragment of DNA were obtained. Among the five clones, SI of C2 and C3, C4 and C5, C1 and C2, and C1 and C4 is 0.993 1, 0.992 7, 0.931 6,and 0.835 7 respectively. These results showed that C1 is a clone, C2 and C3 is a clone, and C4 and C5 is a clone respectively, indicating that the genome of the 4 sporocysts in E. tenella oocyst is different.
