应用花培技术选育水稻光敏核不育系 ①

摘  要:对粳型光敏核不育系,常规粳稻品种及其F1杂种进行花药培养,所得花粉植株用于分期播种和进入人工气候箱以鉴定光敏核不育性.结果表明:不同光敏核不育系及其杂种F1培养力存在明显差异.从组合N5047S/浙农大40和N5047S/02428分别获得84个和35个二倍体花培系,构建成了两个DH群体;在长日下这两个DH群体育性发生明显分离,其中28份为光敏核不育系,短日下各花培系都可育;从N5047S/02428组合中筛选出一个花培系浙农大13S(HP13S),经人工气候箱鉴定表现为光敏核不育特性.不育期为8月上旬到9月2日,长达20多天,制种是安全的,可育期结实率高。The anthers of photoperiod sensitive genic male sterile (PGMS) Japonica rice, convention Japonica rice, and their F 1 hybrids were cultured, and the anther derived lines were used to identify if they had photopoeriod sensitive genic male sterility. The results showed that callus induction frequency,green plant differentiation frequency and anther culture ability had obvious differences among different PGMS lines and different F 1 hybrids. Two DH populations, containing 84 and 35 DH lines respectively, were derived from N5047S/Zhenongda40 and N5047S/02428. The fertility distribution was continuous and the curve of segregation showed two peaks for the both populations under long days. There were 28 anther derived lines which showed photoperiod sensitive genic male sterility. Under short days, all the DH lines wrer fertile. Zhenongda13S (HP13S), an anther derived line, was selected from the cross N5047S/02428. It was a PGMS line by identifying in growth chambers. Its sterile period was more than 20 days from early August to September 2, so it may be reliable to use this line as a sterile parent for hybrid seed production. Moreover its percentage of seed set was high in fertile period, thus seed reproduction was not limited. The results demonstrated that Zhenongda13S was a typic PGMS line.
