摘 要:[目的]建立一种干预体外细胞生长的周期性机械牵拉力学装置。[方法]使用微型电动机控制的支撑物上下位移运动,隆起6孔板具有硅胶弹性的底部基膜,发生周期性形变,使贴附其上的贴壁细胞受牵张应力,并利用三维有限元的方法分析基膜的力学分布,进一步将此装置应用到骨髓间充质干细胞(MSCs)的培养中,对系统进行验证。[结果]此装置操作简单,性能稳定,参数可调性强,生物相容性好,应力变化均匀连续,适当的牵张应力可刺激细胞增殖。[结论]模型可以用于体外应力刺激细胞效应的实验研究。[ Objective] To develop a mechanical stretching-loading device for cell culture. [ Method] A mechanical stretching-loading device for cultivated cells was designed and installed. A bioflex for cell culture dish with a elastic, silicon botton membrane was placed on the support of device. The cell culture plate attached to botton membrane was lyelie stretched and deformed as the menbrane on the support moved up and down controlled by a controlablc micromotor. A three-dimension finite element analysis model was developed to analyze the strain and stress on elastic membrane which represent in turn that on the cultivated cells. Mesencymal stem cells (MSCs) were cultivated on this elastic membrane under a strain rate of 0% ( control group) , 4% (group B) , 6% ( group C) , 9% ( group D), and 12% ( group E) experimental groups. [Result] The device was operated easily and the parameters recoded was adjusted conviniently and reliablly in applying stretching-loading on euhivated cells. As comparison with control group, the cultivated MSCs grew better in a regular arrangement along the lengent direction of applied stress and its proliferation activity was significantly better in group D and group E. [ Conclusion] The new developed tension-loading unit operates easily and provides a reliable parameters of loading tension for cuhivated cells, and can be used to evaluate cell response sensibility to mechanical tension stress.
