摘 要:目的探讨急性肝损伤中肝细胞胀亡与肝组织肿瘤坏死因子-α(TNF-α)的关系。方法应用D-氨基半乳糖(D-GalN)诱导大鼠急性肝损伤模型,同时设立正常对照组,于不同时相检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)含量,肝组织中TNF-αmRNA表达,观察肝细胞胀亡情况,计算胀亡指数(oncosis index,OI)。结果与正常对照组比较,致伤后大鼠血清ALT、AST明显升高(P<0·05),肝组织中TNF-αmRNA表达增强,细胞胀亡增多,OI值增高。结论急性肝损伤中存在细胞胀亡,其发生与TNF-α的表达有一定关系。Objective To investigate the correlation of oncosis and TNF-α in D-galactosamine (D-GalN) induced acute hepatic injury. Methods Acute hepatic injury model was induced by D-GalN in 24 SD rats, and 12 rats served as control. At 4, 8, 12, 16, 20, 24 h after intraperitoneal injection of D-GalN, the levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum, and the tumor necrosis factor-α (TNF-α) in liver tissue were measured. The pathological changes of liver and the oncosis of hepatocytes were observed by TEM. Results In acute hepatic injury, the levels of ALT, AST, TNF-α mRNA, OI were higher than that of control group (P 〈 0.05) , and OI increased with the severity and course of acute hepatic injury. Conclusion Oncosis occurs in acute hepatic injury in rats and its mechanism might be related to the TNF-α expression.
