摘  要:【目的】探讨雷公藤甲素(triptolide A,TA)对健康人外周血单个核细胞(peripheral blood mononuclear cells,PBMC)分泌促炎、抑炎细胞因子的影响。【方法】分离培养健康人PBMC,采用脂多糖(LPS,终浓度为1μg/mL)或LPS+植物血凝素(PHA,终浓度为25μg/mL)刺激培养细胞,并给予TA(终浓度为5.4 ng/mL)处理,收集培养上清液,采用酶联免疫吸附(ELISA)法检测上清液中肿瘤坏死因子α(tumour necrosis factor-,αTNF-α)、白介素4(interleukin-4,IL-4)、白介素10(interleukin-10,IL-10)的含量。【结果】TA能抑制LPS或LPS+PHA刺激的PBMC分泌TNF-α和IL-10,且对正常人PBMC分泌IL-10的抑制作用强于对TNF-α的抑制作用,但对IL-4的分泌无显著性影响。【结论】雷公藤甲素可不同程度地抑制促炎性细胞因子TNF-α及抑炎性细胞因子IL-10,使促炎、抑炎两种作用趋于平衡。[Objective] To investigate the influence of triptolide A (TA) on the production of inflammation-promoting and anti-inflammatory cytokines in peripheral blood mononuclear cells (PBMC) from the healthy volunteers. [ Methods] PBMC isolated from the healthy volunteers were cultured and stimulated by lipopolysaccharide (LPS, the final concentration being 1μg/mL) or LPS + phytohemagglutinin (PHA, the final concentration being 25 μg/mL), and were also treated with or without TA (the final concentration being 5.4 ng/mL) simultaneously. The contents of tumor necrosis factor α (TNF-α), interleukin 4 (IL-4) and interleukin 10 (IL-10) in the supernatant of the culture were measured by enzyme-linked immunosorbent assay (ELISA). [ Results] TA inhibited the production of TNF-α and IL-10 in PBMC culture stimulated by LPS or LPS + PHA, and the inhibitory effect of TA on IL-10 was stronger than on TNF- α, but had no effect on the production of IL-4. [Conclusion] TA has an inhibitory effect on the inflammation-promoting cytokine of TNF-α and anti-inflammatory cytokine of IL-10, which keeps the balance of inflammation-promoting and anti- inflammatory actions.
