摘 要:目的观察侧脑室内注射氟柠檬酸(FCA)对内脏痛大鼠延髓孤束核中即刻早期基因fos蛋白和胶质原纤维酸性蛋白(GFAP)表达的影响,探讨星形胶质细胞和神经元是否参与内脏痛的调节。方法侧脑室置管成功的雄性SD大鼠32只,随机分为4组(n=8):对照组(A组)、内脏痛组(B组)、生理盐水+内脏痛组(C组)和FCA+内脏痛组(D组)。采用腹腔注射0.6%乙酸10 ml·kg^(-1)制备大鼠内脏痛模型。D组侧脑室内注射FCA 10μl(1nmol·μl^(-1))后90 min制备内脏痛模型,C组给予等量生理盐水。观察注射乙酸后60 min内行为学变化,计算内脏痛指数(VPI),注射乙酸后60 min处死大鼠,用荧光免疫组织化学法观察延髓孤束核中fos蛋白和GFAP的表达。结果与A组比较,B组、C组、D组VPI升高,延髓孤束核中fos蛋白和GFAP表达升高(P<0.05);与B组、C组比较,D组VPI降低,延髓孤束核中fos蛋白和GFAP表达降低(P<0.01)。结论延髓孤束核中星形胶质细胞与神经元均参与内脏痛的调节。Objective To investigate the effect of intraeerebro-ventricular (i.c.v.) injection of fluorocitrate (FCA) on expression of los and glial flbrillary acidic protein (GFAP) in nucleus of solitary tract (NST) in medulla oblongata and determine if astroeytes and neurons are involved in the modulation of visceral pain, Methods Adult male SD rats weighing 200-250 g were used in this study. PE-8 tube (internal diameter = 0.2 nm, external diameter = 0.5 nm) was inserted into the lateral ventricle of cerebrum and fixed for i.e.v.. Veseeral pain was induced by intraperitoneal (IP) 0.6% acetic acid 10 ml. kg^-1. Thirty-two animals were randomized into 4 groups ( n = 8 each) : group A did not receive any treatment and served as control ; group B visceral pain; group C normal saline 10 μl i.c.v. + visceral pain and group D FCA 1 nmol in NS 10μl i.e.v. + visceral pain. In group C and D 6% acetic acid was injected IP 90 min after NS and FCA (i.e.v.). The degree of visceral pain was assessed by the number of writhes of the body within 60 min after IP acetic acid and expressed by visceral pain index (VPI) according to Sehmauss (0 = no pain, normal posture, 3 = worst pain, abdominal muscle contraction and abduction of the extended hind limb) . The animals were killed at 60 rain after IP acelic acid injection. The expression of los and GFAP in NST was determined by immunofluorescent method. Results VPI and expression of los and GFAP in NST in medulla oblongata were significantly increased in group B, C and D as compared with group A and were significantly decreased in group D as compared with group B and C, Conclusion Astrocytes and neurons in NST may play an important role in modulation of visceral pain induced by intraperitoneal acetic acid.
