椎间盘突出症大鼠血清白细胞介素-10、白细胞介素-12的变化

摘　　要：目的观察椎间盘突出症大鼠血清白细胞介素-10(IL-10)、IL-12的变化。方法30只雄性SD大鼠,随机分为3组(n=10)：对照组(C组)、实验3d组(T_3组)和实验6d组(T_6组)。T_3组和T_6组硬膜外腔注射20μl自体髓核混悬液和2%利多卡因30μl,C组硬膜外腔注射生理盐水20μl和等量2%利多卡因。T_3组观察3 d,T_6组和C组分别观察6d后剖胸取心室血2 ml,离心后取上清液,酶联免疫吸附法测定血清IL-10、IL-12浓度。结果T_3组和T_6组血清IL-10、IL-12浓度高于C组(P＜0．01或0．05),T_6组IL-10低于T_3组(P＜0．05)。结论IL-10、IL-12在椎间盘突出症发生发展中可能起重要作用。Objective To investigate the changes in serum interleukin-10 （IL-10） and interleukin-12 （IL-12） levels during herniation of nucleus pulposus of intervertebral disc. Methods Thirty adult male SD rats weighing 280-320 g were randomly divided into 3 groups（ n = 10 each） ： group Ⅰ control; group Ⅱ 3 day experiment and group Ⅲ 6 day experiment. The animals were anesthetized with intraperitoneal 1% ketamine 50 mg · kg^ - 1 . Five nuclei pulposus of coccygeal vertebra were obtained from each animal in group Ⅱ and Ⅲ and added to 50 μl of normal saline （NS） and made into autologous nucleus pulposus suspension. In group INS 20μl and 2% lidocalne 30 μl were injected into epidural space at L4,5 interspace and the animals were observed for 6 days. In group Ⅱ and Ⅲ autologous nucleus suspension 20 μl and 2% lidocaine 30 μl were injected into epidural space and the animals were observed for 3 and 6 days respectively. Blood samples were taken from the ventricle of heart at the end of 3 days （group Ⅱ ） or 6 days （group Ⅰ ,Ⅲ ） for measurement of serum concentration of IL-10 and IL-12 by ELISA.Results The serum IL-10 and IL-12 concentration were significantly higher in group Ⅱ and Ⅲ than in group Ⅰ , while the serum IL-10 concentration in group Ⅲ was significantly lower than that in group Ⅱ （ P 〈 0.05 ）. Conclusion IL-10 and IL-12 may play an important role in the syndrome of intervertebral disc.
