甘草、桔梗总皂苷对体外磷脂酶A_2活性的影响

摘　　要：目的:建立一种体外磷脂酶A2活性测定的方法,并观察甘草总皂苷、桔梗总皂苷体外对磷脂酶A2活性的影响。方法:以指示剂法为基础,将反应转移至96孔微孔板,用酶标仪在492nm处测定吸收值,以判定药物对PLA2活性的影响。结果:在一定范围内,酶浓度、反应时间与pH值呈良好线性关系。甘草皂苷和桔梗皂苷对PLA2活性有明显抑制作用,其IC50分别是:0.784mg/m l和1.824mg/m。Objective： To improve a method for the determination of the phospholipase A2 activity, the effects of glycyrrhizin and platycodin on phosphohpase A2 activity in vitro are observed. Methods： Based on the absorbance changes of indicators with the concentration of hydrogen ion released from an enzyme-catalyzed reaction, a convenient colorimetric method for the determination of phospholipase A2 activity was modified, and all tests were performed in microplates. By measuring the OD values of microplates in 492nm, The effects of drugs on phospholipase A2 activity were evaluated. Results ： In a certain range of the enzyme concentration and reaction time, there is a well linear relationship between them and pH value. Both glycyrrhizin and platycodin can obviously inhibit the phospholipase A2 activity, the IC50 value of glycyrrhizin on inhibiting the phospholipase A2 activity is 0. 784mg/ml and that of platycodin is 1. 824mg/ml.
