摘  要:目的对混和于宿主蛋白的约氏疟原虫红外期蛋白进行初步研究。方法制备针对疟原虫唾液腺子孢子的免疫血清,通过免疫印迹识别肝细胞内疟原虫裂殖体的相关蛋白,并对识别的蛋白进行肽指纹图谱分析。结果通过免疫印迹成功地识别了疟原虫相关的蛋白,经MALDI-TOF-MS获得肽指纹图谱,以Mascot软件分析并在疟原虫蛋白数据库中检索到了相匹配的多肽。结论应用免疫印迹与肽指纹图谱相结合的方法,可以对存在于复杂混和物中的红外期疟原虫蛋白进行初步分析。Objective To detect and analyze the Plasmodium yoelii liver stage proteins in rat liver extracts, Methods Immune sera were prepared by immunizing rabbits with lyzed salivary sporozoites, Liver stage proteins were analyzed by Western-blot. Protein bands in the SDS-PAGE gel, after staining with coommassie brilliant blue, were collected and analyzed by MALDI-TOF-MS. The results obtained from peptide mass fingerprint (PMF) were analyzed by Mascot search program (http://www,matrixscience,com/). Results Liver stage proteins of P. yoelii were successfully identified. Conclusion The liver stage plasmodium proteins in rat liver extracts can be analyzed by Western-blot and MALDI-TOF-MS.
