摘　　要：目的研究可溶性速殖子抗原(solubletachyzoiteantigen,STAg)和霍乱毒素(choleratoxin,CT)佐剂滴鼻免疫BALB/c小鼠后鼻相关淋巴组织(nasal-associatedlymphoidtissue,NALT)的免疫效应及持续时间。方法BALB/c小鼠96只随机分为实验组和对照组,实验组以免疫原性好的STAg(20μg/只)为抗原,CT(1μg/只)为佐剂滴鼻免疫,对照组以PBS滴鼻。滴鼻2次(间隔2周)后分别于第1、2、3、4、6、8、10、12周处死6只小鼠。分离NALT的淋巴细胞,计数并涂片,用免疫细胞化学法(immunocytochemistry,ICC)检测其CD4+、CD8+T细胞亚群水平。结果小鼠免疫后,NALT淋巴细胞第1、2、6、8、12(P<0.05)周显著增生,可持续至免疫后第12周;CD4+T细胞第1周至第6周(P<0.05)持续显著增高,CD8+T细胞在第1周至第4周持续增高(P<0.01),CD4+/CD8+比值第2、3周显著降低(P<0.05)。结论STAg和CT佐剂滴鼻免疫BALB/c小鼠能够有效诱导NALT的免疫效应,并可持续较长时间。Objective To study the cell-mediated immune response in nasal-associated lymphoid tissue （NALT） after intranasal immunization with STAg and CT adjuvant. Methods Ninety six BALB/c mice were randomly divided into two groups ： For the experimental group, mice were intranasally immunized with 20 μg STAg and 1 μg CT twice at an internal of two weeks. For the control group, mice were given with PBS. Group of six mice from each group were killed at week-1,2,3,4,6,8,10, and 12 after the last immunization. Lymphocytes in NALT were isolated and the percentages of CD4^＋ and CD8^＋ T-cell were determined by ICC. Results The total number of T-cell was found to increase at week-1 （P〈0.01）, 2 （P〈0.05）,6 （P〈0.001）, 8 and 12 （P〈0.05）. The level of CD4^＋ T-cell increased significantly at week-1, 2, 3 （P〈0.001）, 4 and 6 （P〈0.05）, and the levels of CD8^＋ T-cell in the immunized mice （week- 1, 2, 3, and 4） were higher than the control mice. The ratio of CD4^＋ to CD8^＋ T-cell decreased at week-2 and 3 （P〈0.05）. Conclusion Intranasal immunization of mice with STAg and CT adjuvant can effectively induce a long lasting cellmediated immune response in NALT.
