摘 要:目的:探讨左归丸促进骨髓形成肝细胞的分子机制。方法:采用交叉性别骨髓移植模型,雌性BALB/c小鼠接受9.0Gy60Co源γ射线照射后,经尾静脉注射输入同种系雄性小鼠骨髓细胞,分左归丸组和模型组,6个月后处死实验动物,取肝组织标本。选用小鼠基因表达谱寡核苷酸芯片,观察实验小鼠肝组织基因表达谱的变化。结果:左归丸组小鼠在骨髓移植6个月后肝组织的基因表达谱与模型组比较有显著不同,差异表达基因有1147条,已知功能基因有533条,其中上调基因264条,下调基因269条。结论:左归丸可能是通过影响肝组织基因表达谱的变化促进骨髓形成肝细胞。Objective:To study the molecular mechanism of Zuoguiwan in promoting bone marrow cells to form hepatic cells. Methods:After irradiation with 9.0Gy ^60Co Y-rays,the female BALB/c mice were injected via the tail vein with bone marrow cells of homogenic male mice to establish bone marrow transplantation model. The female BALB/C mice were randomly divided into a Zuoguiwan group,and a model group. After 6 month,all mice were sacrificed and liver tissues were taken. Changes ence of gene expression profile was detected using the mouse gene expression profile oligo-nucleotide chip. Results: There were significant differences in gene expression profile of the liver tissue between the two groups after 6 months of transplantation ,with 1 147 genes of differential expression,of which 533 genes were function-known and 264 genes were up-regulated, 269 genes down-regulated. Conclusion: The molecular mechanism of Zuoguiwan in promoting bone marrow cells to form hepatic cells is exerted possibly via influencing the changes of gene expression profile.
