摘 要:以从4个草坪型高羊茅品种成熟种子直接诱导获得的初始愈伤组织为起始材料,研究3种因素对胚性愈伤组织继代发生的影响。结果表明,继代培养基中添加BAP和硫酸铜以及提高蔗糖浓度均能促进胚性愈伤组织形成,提高胚性愈伤组织频率,其适宜浓度分别为:BAP0.1~0.2mg/L,硫酸铜2.5mg/L,蔗糖60g/L。这3种因素中,又以添加BAP的效果最好,添加硫酸铜次之。4个供试品种间胚性愈伤组织发生能力存在明显差异。The effects of three different factors on subculture production of embryogenic calli were investigated with ones induced directly from mature seeds of four tuff-type tall fescue cultivars as starting materials. The results showed that supplementing BAP and cupric sulphate and increasing sucrose concentration in subculture medium stimulated formation of embryogenic calli, with appropriate concentration being 0.1 - 0.2 mg/L for BAP, 2.5 mg/L for cupric sulphate, and 60 g/L for sucrose. Among these three factors, BAP addition was the most effective, followed by supplementation of cupric sulphate. It was also demonstrated that apparent aifferences in productivity of embryogenic callus existed among the four tested cultivats.
