摘 要:利用选择性培养基从发霉的豆粕、豆油工厂周围环境中取样并分离纯化获得一株产热稳定性α-半乳糖苷酶耐热真菌,通过形态学和分子生物学鉴定方法初步鉴定该菌株为分枝犁头霉.研究表明该菌株生长的最适温度和pH值是47℃和7.0,所产酶为胞内酶.酶活研究表明粗酶液的最适温度和pH值分别为68℃和7.0,具有较好的热稳定性.A thermotolerant fungus was isolated from the soil sample collected near the factory which produces soybean oil by using a selective medium. A strain capable of producing thermostable α - galactosidase was identified as an Absidia Ramose WL511 according to the morphological and the molecular characterization. The optimal conditions for the growth of the strain were examined. The optimal temperature and pH value for growth were 47℃ and 7.0, respectively. Research on the activity of the α - galactosidase showed that the optimal temperature and pH for the activity of the crude enzyme was 68℃ and 7.0, respectively. The α- galactosidase from Absidia Ramose WLSll was located intracellularly.
