摘  要:通过对玉米DUS测试技术体系构建的研究,结果为:①确定了49个品种组成的玉米DUS测试标准品种群体,补充和完善了原来的群体;②49个DUS测试性状的差异指数为0—8.00,性状20差异指数最大,有30个性状的差异指数为0~2.00,表现较稳定;③26个参试标准品种的差异指数为1.89—8.11,中451最小,掖52106最大;④用方法1得到的DNA的A260IA230为2.119,A260/A280值为1.814,说明方法1所得的DNA纯度高、浓度高、条带清晰、降解少、质量优。⑤从国内、外所用的63对核心引物中筛选出PIC值高,条带清晰,稳定性好的20对玉米SSR核心引物;⑥用8%聚丙烯酰胺电泳在20对引物扩增产物中检测出了171个位点,PIC值为0.8554,远远高于3%琼脂糖电泳,更适合DUS测试;⑦探讨了玉米DUS快速测试技术的建立方法。This paper studied the establistunent of technology systerm for DUS test in Maize.The results showed that ( Ⅱ )A group with 49 maize starndard varieties for DUS test,which it was better than old one,have been built. ( Ⅱ )Among 0 - 8.00, the differential indexes (DI) of 49 DUS testing characters were analyzed. DI of No. 20^th character was the most biggest, and the DI which the 30 DUS testing characters with higher stable less than 2.00. (Ⅲ )The variation of DI from 26 stardand varieties for DUS test were among 1.89 - 8.11. DI of oAtivar Zhong 451 was the least, and Ye 52106 was the most biggest. ( Ⅳ ) Purity (A260/A280 = 1.814; A260/A230 = 2.119) and quality of maize DNA by 1 st methed was better than it by another 2 metheds; ( V ) 20 pairs core primers out of 63 pairs of core SSR primers which had were used by other labs were selected which gave stable and high polymorphism infomation .content (PIC) ; ( Ⅵ) 8 % polyacrylamide gel was fitter for DUS testing than 3 % Metaphor agarose gel. Among the same amplification yield, 171 alleles and higher PIC value (0.8554) were detected by 8 % polyacrylamide gel which was over higher than it by 3 % Metaphor agarose gel; (Ⅶ)Based on above,we discussed the building of fast methed for DUS testing in new maize cultivars.
