摘 要:目的探讨HCK(hemopoietic cell kinase)基因在心房肌组织中的表达与房间隔缺损(ASD)的病理生理学关系。方法获取10例单纯Ⅱ孔型ASD患者和8例正常对照者右房心耳肌组织标本,采用RT-PCR技术检测HCK基因mRNA的表达丰度。结果HCK基因mRNA在正常心房肌和ASD心房肌组织中均有表达,但ASD患者心房肌组织中的HCK基因表达丰度为(24.50±10.29)%,显著低于正常对照组(60.19±17.03)%(t=5.5100,P<0.01)。结论HCK基因在心房肌组织中的异常表达可能与ASD的病理生理学相关。Objective To explore the changes of hemopoietic cell kinase (HCK) gene mRNA expression in human auricular myocardium of atrial septal defect (ASD) and analyze the relationship between abnormally expression of HCK gene and the pathophysiology of ASD, Methods The total RNAs were extracted from 10 cases of ASD auricular myocardium and 8 cases of normal auricular myocardium. The levels of HCK gene mRNA expression in the auricular myocardium of ASD and the normal auricular myocardium were detected using RT-PCR technique. Results The HCK gene expressed in both the normal and ASD auricular myocardium. The level of HCK gene mRNA expression was 24.50%± 10. 29% in the ASD auricular myocardium, which was significantly lower than that 60. 19% ± 17. 03% in the normal auricular myocardium (P〈 0.01 ). Conclusion The abnormally expression of HCK gene in the auricular myocardium may contribute to the pathophysiology of ASD.
