摘  要:目的探讨外源性神经生长因子(NGF)对急性脊髓伤后运动神经功能恢复的影响。方法SD大鼠40只,随机分为对照组20只、NGF组20只。Allen’s的WD法,造成T8节段急性脊髓挫伤。NGF组于伤后1周内通过微泵将NGF经置入皮下的导管泵入蛛网膜下腔。分别于麻醉清醒后、1周末、4周末,采用Gale联合计分法(CBS),计算脊髓损伤程度的百分数。4周末,以辣根过氧化物酶(HRP)逆行示踪脊髓下行传导通路,计算阳性细胞数及面积。结果损伤4周后CBS记分及HRP逆行示踪提示NGF组与对照组比较运动神经功能恢复有显著性差异(P<0.01)。结论持续外源性NGF可促进脊髓损伤后运动神经功能的恢复及神经通路的重建。Objective To study the effect of exogenous nerve growth factor(NGF) on neurological function of motor nerve after expreimental spinal cord injury. Methods Fourty SD rats were randomly divided into control group and NGF group. Allen's weight-drop(WD) technique with 25 gcm (10 g×2.5 cm) force onto the exposed dura at T8 was carried out to produce contusion injury(SCI). NGF was pumped via subarachnoid space route 1 week after SCL Neurological function was assessed by Gale' s combined behavioral score(CBS) on the 1st day,at the end of 1 week and 4 weeks. 4 weeks after injury, 36 animals were retrogradely labelled to show rubrospinal neurons by horseradish peroxidase(HRP) ,and the labelled cell number and area were calculated. Results Four weeks after injury, CBS and the labelled cell number and area were markedly bighter in NGF group than those in control group(P〈0.01). Conclusion Exogenous NGF pumping via subarachnoid space may promote the recovery of motor nerve and the reconstruction of nerve path after experimental spinal cord injury.
