摘 要:目的:观察唐古特大黄多糖(rheum tanguticumpolysaccharides,RTP)对小鼠溃疡性结肠炎(ulcerativecolitis,UC)的疗效并探讨其可能存在的机制。方法:应用三硝基苯磺酸(TNBS)灌肠制备小鼠溃疡性结肠炎模型。实验设正常对照组、模型对照组、RTP治疗组(100、200、400 mg.kg-1)。观察各组实验小鼠体质量变化及组织病理学改变,采用酶联免疫吸附法检测细胞因子(IL-8、IL-10、TNFα-、IL-4)的含量。结果:中、高剂量的RTP能够缓解小鼠体质量的减轻,减少腹泻及便血的发生;并且能够显著改善小鼠结肠组织损伤及病理学改变。溃疡性结肠炎模型组TNFα-、IL-8的表达较正常对照组显著增高,而IL-10、IL-4的表达明显降低(P<0.05);高剂量RTP治疗组的TNFα-、IL-8表达明显低于溃疡性结肠炎模型组,IL-10、IL-4的表达显著高于模型组(P<0.01)。结论:RTP对TNBS诱导的小鼠溃疡性结肠炎具有良好治疗作用,其机制可能与上调抗炎细胞因子(IL-10、IL-4)及下调促炎细胞因子有关。AIM: To explore the effects of rheum tanguticum polysaccharides (RTP) on TNBS -induced colitis in mice and its probable mechanisms. METHODS: Mice colitis model was induced by 2, 4, 6-trinitrobenzene sulfonic acid (TNBS). The mice were randomly divided into 5 groups,normal group, model group, RTP-100, 200 and 400 group. The maeroseopieal and histological changes of the colon were evaluated and the eytokines IL-8, IL- 10, TNF-α and IL-4 produced by splenoeyts were analyzed with ELISA. RESULTS: Compared with the model group, both symptoms and the lesions of colonic mueosa of RTP-200 and 400 group were slighter on ulcerative coliris induced by TNBS in mice. Furthermore, the expressions of TNF-aand IL-8 in colon tissue of mice with TN- BS-induced colitis were higher and the IL-10 and IL-4 were lower than that of normal control ( P 〈 0.05 ). In RTP-400 group, the positive expressions of TNF-α and IL-8 were lower and IL-10 and IL-4 were higher than that of model group( P 〈 0.01 ). CONCLUSION: RTP has significant effects on TNBS-induced colitis in mice, the mechanism of which may relate to its regulation on cytokines.
