摘 要:目的研究甲氨喋呤磁靶向脂质体在小鼠体内的组织分布。方法建立了高效液相色谱法测定给药后小鼠体内血浆和组织中的药物浓度;采用逆向蒸发法自制了甲氨喋呤磁靶向脂质体;研究了其在体内的组织分布和磁靶向作用。结果所建立的HPLC线性良好,回收率高,精密度好;经过小鼠尾静脉给药后,在两肾脏处设一磁场,结果证明,具有很好的靶向性。结论甲氨喋呤磁靶向脂质体磁场条件下在靶部位有明显的浓集,具有良好的靶向性,所建HPLC用于甲氨喋呤体内分布结果良好。OBJECTIVE To study on the tissue distribution of methotrexate magnetoliposomes in mice after an intravemons administration. METHODS A HPLC method was established to determine the concentration of MTX in plasma and tissues. The liposomes were prepared by the reverse evaporation method. Tissue distributions of methotrexate magnetoliposomes and magnetic targeting were studied. RESULTS The established method was accurate, replicable and with good linearity. After an administration, the methotrexate magnetoliposomes were concentrated in the area where the magnetic field was applied. CONCLUSION The methotrexate magnetoliposomes show the good targeting property when being exposed in the magnetic field. The established HPLC method is easy to be used in MTX biodistribution study.
