尿激酶热敏脂质体溶栓作用的体外实验研究

摘　　要：目的:探讨尿激酶热敏脂质体(UK-TsL)的体外热靶向溶栓能力。方法:32个血凝块随机分为对照组、游离尿激酶(FUK)组、UK-TsL组和尿激酶脂质体(UK-L)组。将血凝块置于附有加热装置的简易体外溶栓装置中,分别加入上述4组试剂,从输液开始对血凝块以42℃或37℃的温度加热,输液120min后测定血凝块的溶解率。结果:UK-TsL组血凝块溶解率达50.41%,与FUK组相近;与对照组及UK-L组相比有显著性差异(P<0.01)。结论:UK-TsL在42℃时迅速释放尿激酶,具有较好的热靶向溶栓能力,能有效减少药物剂量。Objective: To explore the value of urokinase encapsulated thermosensitive liposome(UK-TsL) in thermally targeted thrombolysis.Method: 32 blood clots were divided randomly into control group,free urokinase(FUk) group,UK-TsL group and urokinase encapsulated liposome(UK-L) group.The blood clots were put into a simple vitro-thrombolysis-equipment with heating instrument,then the four reagents upword were added respectively.The blood clots were heated at 42℃ or 37℃.120 minutes after transfusion,the thrombolytic rate of blood clots was measured.Results: The average thrombolytic rate of blood clots was up to 50.41 in UK-TsL group,which was close to FUK group.Significant differences were observed among UK-TsL group,control group and UK-L group (P<0.01).Conclusion: UK-TsL can quickly release urokinase at 42℃.It has the thermally targeting property in clot thrombolysis,which can reduce the dosage of medicine efficiently.
