摘　　要：目的:通过对大鼠骨髓间充质干细胞(BMSCs)转染人TGF-β1基因,观察该基因对BMSCs表达I型胶原的影响。方法:采用脂质体介导法转染人TGF-β1基因至BMSCs,采用定量RT-PCR,免疫荧光分析等方法分析转基因后,BMSC表达I型胶原的变化。结果:重组质粒pCDNA3.1(+)-TGF-β1成功转染至大鼠BMSCs,细胞免疫荧光和定量RT-PCR均证实了TGF-β1表达的增强。结论:转TGF-B1基因可以促使BMSCs合成I型胶原。Objective：To study the effect of transforming growth factor-β1 （TGF-β1）on protein expression of collagen Ⅰ of BMSCs by transfected human TGF-β1 gene. Method： Human TGF-β1 gene was transferred into BMSCs by liposome and confirmed by RT-PCR.Result：Reconstructed plasmid pEDNA3.1 （＋）-TGF-β1 was successfully transfected into BMSCs, RT-PCR showed the protein of collagen Ⅰ was highly expressed in transfected cells （P〈0.01）. Immunofluorescence analysis showed that the expression of collagen Ⅰ in transfected cells was much higher than that in the control group. Conclusion： Transfecting human TGF-β1 gene into BMSCs could promote the expression of type Ⅰ collagen.
