摘 要:研究了在pH9.5的NH3·H2O-NH4Cl缓冲溶液中,DNA对5-(8-喹啉偶氮)-罗丹宁(QARH)的比率不同,DNA与QARH形成3种逐级复合物。第1级复合物的λmax为440nm,DNA的最大结合数为30;第2级复合物的A~为460nm,DNA的最大结合数为111;第3级复合物的λmax为490nm,DNA的最大结合数为645。DNA与QARH的第l级结合服从Pesavento模型。提出了用QARH测定DNA的方法,DNA含量在1.0~50.0mg/L范围内服从Beer定律,所提出的方法可用于植物DNA提取过程中DNA产率的鉴测。The interaction between DNA and 5- ( 8-quinolylazo)- rhodanine(QARH) was studied. In a NH3. H2O-NH4CI buffer solution with pH 9.5, DNA could form three complexes with QARH while the ratio between DNA and QARI-I was changed. The λmax of the first, second and third complexes was 440,460 and 490 nm respectively, and the maximum binding number of DNA in the complexes was 30, 111 and 645 respectively. The model of the first combination of DNA with QARI-I obeyed the Pesavento model. A method for the determination of DNA was proposed using QARH and was used to determine DNA in the solution for extracting the plant DNA. The Beer's law was obeyed when the DNA concentration ranged from 1.0 mg/L to 50 mg/L of DNA.
