摘 要:为提供急性心肌梗塞(AMI)早期血清学诊断法,用改进后的 Sch βler 法提取狗肌凝蛋白轻链(CM-LC),免疫兔获得抗血清。氯胺 T 法标记抗原,建立血清 CM-LC RIA。结果:该方法标准曲线测量范围1~100μg/L,批内、批间 CV 分别是3.2%和9.1%,平均回收率95.3%。正常人30例血清 CM-LC 为3.5±1.5μg/L,20例 AMI 患者血清 CM-LC 为20.2±8.6μg/L,两组间差异非常显著。提示血清 CM-LC RIA 可用于 AMI 早期诊断。PURPOSE To establish a radioimmunoassay (RIA) of cardiac myosin light chains (CM-LC) for early diagnosis of acute myocardial infarction (AMI).METHODS The method established by Sch(?)βbler was modified to purify CM-LC from fresh dog heart.Antiserum was obtained by immunizing rabbits.CM-LC was iodinated by chloramine-T method.Conventional RIA procedure was used.RESULTS Sensitivity of the RIA system was 1μg/L.The intra-assay CV and inter-assay CV were 3.2% and 9.1% respectively,and the mean recovery rate was 95.3%.The serum CM-LC level in 30 normal subjects was 3.5±1.5μg/L, while in 20 patients with AMI was significantly increased (20.2±8.6/μg/L,P<0.01).Increased CL-MC concentration in pa- tients was seen at 5 to 72 hours after the attack of AMI.CONCLUSIONS These results suggest that RIA of CM-LC could be valu- able in the diagnosis of AMI.
