摘  要:本文建立了测定家兔血浆中Z-47(3H-1,2-二氢-2-(4-甲基苯胺基)甲基-1-吡咯里嗪酮)的高效液相色谱法。选用μ-BondapakC18色谱柱,流动相为甲醇-磷酸二氢铵缓冲液(pH=4)(3:2,V/V)。以安定为内标物。血浆样品先经碱化,用乙醚-正己烷(1:1,V/V)提取,分离有机相,再以10%磷酸萃取,水相20μL进样,紫外检测波长290nm。本法专属性强,精密度、准确性符合有关要求,操作简便。对6只家兔口服给药后应用本法成功地进行了初步的药代动力学研究。A high-performance liquid chromatographic method for the quantitative determination of Z-47(3H-1,2-dihydro-2-(4-methylphenylamino)methtl-1-pyrrolizinone),a new antiinflammatory and analgesic agent,in rabbit plasma is presented.The method uses a μ-Bondapak C18 column and a mobile phase containing methanol and ammonium dihydrogen phosphate buffer(pH4)(3:2),with diazepam as the internal standard.The analyte is detected by UV at 290 nm.Calibration curves are linear over the range of 30-3000 μg/L.The limit of quantitation is 30 ng/mL for Z-47 in the plasma.The method proves to be simple and sensitive with good specificity,precision and accuracy.It has been successfully applied to a preliminary study of Z-47 pharmacokinetics involving six rabbits.The plasma samples have been alkalified and then extracted with ether-n-hexane(1:1).The organic phase has been separated and extracted with 10% phosphoric acid subsequently.20 μL of the aqueous phase has been injected into the chromatographic system.
