摘  要:目的 探索电致孔条件下不同电学参数(电压、波形、脉冲宽度、脉冲次数)对青藤碱透皮给药的影响。方法用中药电致孔给药仪产生多种电学参数条件,并以双室扩散池法、HPLC法检测青藤碱透过小鼠腹部皮肤的变化。结果青藤碱的累积透过量与电参数密切相关,在单因素影响中,4个最佳电学参数分别为:600V电压、正弦波+方波组合波形、500μs的脉冲宽度、240个脉冲次数。透过量有极显著的增大,与被动扩散相比增加幅度有5~100倍。结论 优化的电致孔条件应用于中药有效成分透皮给药有良好的前景。Objective To observe the effects of different electrical parameters (voltage, snap ol wave, pulse width, and number of pulses) on transdermal delivery of sinomenine using eletroporation. Methods The lransdermal penetration in Franz diffusion cell for isolated rat skin of sinomenine was studied by dif ferent electrical parameters of electroporation equipment for Chinese materia medica, respectively. The content of sinomenine per-meated abdomen skin of mice was determined by HPLC. Results The accumulative permeation of sinomenine was correlated with electrical parameters, different electrical parameters showed the significant effect on sinomenine's transdermal delivery. The optimum electrical parameters were voltage: 600 V, shap of wave: the combination of square wave and sine wave, pulse width: 500μs, number of pulses: 240. The permeation increased significantly and the increasing extent was 5-100 times compared with the passive diffusion. Conclusion Optimum parameters of eletroporation have an favorable future for active constituents in Chinese materia medlca to be transdermically deliveried.
