摘 要:目的证实外源性层粘蛋白能否促进复方壳多糖组织工程皮肤基底膜的构建。方法在体外培养组织工程皮肤的培养基中加入浓度为0、1、5、20μg/ml的外源性层粘蛋白,培养15d,通过HE染色及PAS染色,观察获得的组织工程皮肤的组织学特性;并利用免疫组化染色对基底膜的重要成分包括Ⅳ型胶原和层粘蛋白进行观察,观察基底膜构建情况。结果加入20μg/ml的层粘蛋白,可明显促进基底膜的形成,且免疫组化染色结果显示Ⅳ型胶原和层粘蛋白反应强阳性。结论层粘蛋白可促进复方壳多糖组织工程皮肤基底膜的构建,并呈浓度依赖性。Objective To prove the possibility of exogenous laminins to facilitate the basement membrane (BM) formation in tissue-engineered skin. Methods Laminins at the concentrations of 0, 1,5 and 20 μg/ml respectively was added into the keratinocyte growth medium to prepare composite chitosan tissue-engineered skin. After 15 d, the histological manifestation of the skin was observed by HE and PAS staining, while cytokeratin 14 (CK14), collagen Ⅳ ( COL-Ⅳ ) and laminins(LN) at the dermal-epidermal junction were detected with immunohistochemical assays. The formation of basement membrane was assessed. Results The BM in the medium with 20μg/ml laminins developed significantly better than other groups. Immunohistochemical studies showed strong COL-Ⅳ and LN at the dermal-epidermal junction in the tissue-engineered skin. Conclusion Laminins may facilitate the formation of basement membrane in composite chitosan tissue-engineered skin in a concentration-dependent fashion.
