摘 要:本实验观察了胰岛素样生长因子Ⅱ(IGF-Ⅱ)对大鼠离体培养黄体细胞孕酮生成的影响,并对其作用机制进行了探讨。结果显示,IGF-Ⅱ能显著地促进大鼠离体培养黄体细胞孕酮生成并呈剂量-效应关系,同时还能促进3H-亮氨酸(3H-len)掺入黄体细胞蛋白质的合成,促进3H-胸腺嘧啶(3H-TdR)掺入DNA的合成,而上述效应分别被放线菌酮(CYX)和放线菌素D(ActD)所抑制。此外,IGF-Ⅱ对大鼠高体黄体细胞内源性cAMP和hCG诱导的cAMP含量均无影响,这提示IGF-Ⅱ对大鼠黄体细胞孕酮生成的促进作用与其增强黄体细胞蛋白质与DNA的合成有关,而其跨膜信息传递似不通过cAMP系统。It has been reported recently that IGF- Ⅱplays an important role in the regulationof ovarian function. The aim of this study is to investigate whether IGF-Ⅱ can affectprogesterone production by cultured rat luteal cells. The results showed that IGF-Ⅱcould significantly stimulate basal progesterone production in a dose-dependant mannerwithout altering the cAMP content. IGF-Ⅱ could enhance 3H-len incorporation into protein and 3H-TdR incorporation into DNA of the luteal cells, but these effects could beinhibited by CYX and Act D. The above results suggest that stimulating effect of IGF Ⅱ on progesterone production by rat luteal cells may be related to enhanced protein andDNA synthesis.
