摘  要:目的:对野生和栽培藏药麻花艽中龙胆苦苷、落干酸、獐牙菜苦苷和獐牙菜苷4种苦苷类成分进行高效液相色谱的含量测定,并比较分析它们之间的差异。方法:采用 Eclipse XDB-C_8色谱柱(4.6mm×150mm,5μm),流动相 A 为95%乙腈水溶液,B 为5%乙腈(含10mmol·L^(-1)的甲酸)水溶液,A 在0~20min 内比例由0→100%进行线性洗脱,流速1.0mL·min^(-1),检测波长240nm,柱温30℃。结果:4种成分均达到基线分离,龙胆苦苷、落干酸、獐牙菜苦苷、獐牙菜苷的线性范围分别为0.60~19.20μg(r=0.9999),0.24~7.68μg(r=0.9999),0.38~12.02μg(r=0.9999),0.05~1.66μg(r=0.9999);回收率分别为99.73%,98.13%,98.45%,96.22%。结论:栽培藏药麻花艽中苦苷类成分的含量已经接近或超过野生种的水平,可初步代替野生药材入药。Objective:To determinate the contents of four iridoids glycosides, gentiopicroside, loganic acid, swertiamarin and sweroside in wild and cultivated Gentiana straminea Maxim. by HPLC. Method:The samples were separated on an Eclipse XDB - Ca column(4.6 mm × 150 mm,5 μm) with 95% acetonitrile (A) and 5% acetonitrile (10 mmol · L^-1 formic acid) (B) gradient elution with flow rate at 1.0 mL · min^-1. Mobile phase(A) increased from 0 to 100% in 20 min with UV detection at 240 nm and column temperature at 30 ℃. Results: Four compounds were isolated on base line. The linear ranges of iridoids, gentiopicroside, loganic acid, swertiamarin and sweroside were 0.60 - 19.20 μg(r =0. 9999) ,0. 24 -7.68 μg(r =0. 9999) ,0. 38 - 12.02 μg(r =0. 9999) ,0.05 - 1.66 μg( r = 0. 9999 ) , respectively. The average recoveries were 99.73 % , 98.13% , 98.45 % and 96. 22% , respectively. Conclusions: The contents of four iridoid glycosides in cultivated Gentiana straminea have reached or exceeded those in the wild one. Therefore,it is preliminarily feasible for the cultivated G. straminea used as a herbal medicine to replace the wild one.
