摘  要:目的探讨异丙酚预处理对全脑缺血再灌注大鼠脑组织S100β及神经元特异性烯醇化酶(NSE)含量的影响,评价异丙酚对脑组织的保护作用。方法30只雄性SD大鼠,随机分为假手术组(A组)、单纯脑缺血再灌注组(B组)和缺血前2h异丙酚预处理组(C组),每组10只,C组动物在缺血前腹腔注射异丙酚100mg/kg,采用线栓法制作全脑缺血再灌注模型,于脑缺血再灌注后24h评估并记录动物的神经行为学评分,测定大鼠脑组织S100β和NSE含量。结果异丙酚预处理组神经行为学评分较单纯脑缺血再灌注组高(P<0·05),假手术组评分也明显高于单纯脑缺血再灌注组(P<0·05),异丙酚预处理组脑组织中S100β和NSE含量明显低于单纯脑缺血再灌注组(P<0·05)。结论异丙酚可以降低脑缺血再灌注损伤大鼠脑组织S100β和NSE的含量,减轻神经损害,缺血前2h异丙酚预处理可以改善大鼠脑缺血再灌注损伤后的神经行为学评分,有明显的脑保护作用。Objective To study the effects of propofol pretreatment on S100β and enzyme (NSE) of brain tissues with global cerebral ischemia-reperfusion in rat, and to evaluate the effects of propofol in protection of brain. Methods 30 SD male rats were randomly divided into 3 groups: sham operation group (group A, n = 10); single cerebral ischemia-reperfusion group (group B, n = 10); propofol pretreatment at 2h before ischemia group (group C, n = 10), in which propofol (100mg/kg) was given intraperitoneally (ip) before ischemia. 24h after ischernia-reperfusion, the neuroethology scores were recorded and evaluated, and S100β and NSE in rat brain were determined. Results The nettroetbology scores of group A were higher than those of group B (P〈0.05), and the scores of group C were also significantly higher than those of group B (P〈0.05). The S100β and NSE in the rats of group C were significantly lower compared to those of group B (P〈0.05). Conclusion Propofol could reduce S100β and NSE in rat's brain with global cerebral ischemia-reperfusion, and alleviate neural lesions. The neuroethology scores may be improved, and a promising brain-protective effect may be achieved with propofol pretreatment at 2h before ischemia.
