摘 要:目的观察灵芝多糖(GLP)对阿尔茨海默病(AD)大鼠海马组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量以及空间学习记忆能力等的影响。方法双侧海马内一次性注射β-淀粉样多肽25~35片段(Aβ25-35)制作大鼠AD模型,腹腔注射灵芝多糖水溶液,1周后进行Morris水迷宫检测大鼠空间学习记忆能力变化。采用分光光度法测定大鼠海马组织SOD活性和MDA含量。采用透射电镜观察海马神经元的超微结构变化。以及灵芝多糖对上述各指标的影响。结果灵芝多糖能明显改善AD模型大鼠低下的空间学习记忆能力。显著提高模型大鼠海马组织SOD活性及降低MDA含量,而且灵芝多糖能明显改善模型大鼠脑组织海马CA1区神经元的退行性变化。结论灵芝多糖能提高海马内抗氧化酶SOD活性,降低丙二醛MDA含量,改善海马CA1区神经元的退行性变化。对老年性痴呆大鼠学习记忆能力可能有增强和提高作用。Objective To observe the effects and mechanism of Ganoderma Lucidum Polysaccharides (GLP) on the activity of superoxide dismutase( SOD), the content of malondialdehyde(MDA) of hippocampus tissue and the learning and memory ability. Methods AD rats induced by Aβ25- 35 were treated with GLP for 7 days and then observing the Morris water maze to study the learning and memory ability, the content of MDA, and the activity of SOD was measured in the hippocampus tissue with spectrophotometer. Ultrastructural changes of neuron were viewed under transmission electron microscope. Results GLP can distinctly improve the learning and memory ability of Aβ-injected rats( P 〈 0.05), markedly improve the activity of SOD and reduce the content of malondialdehyde(MDA) .The number and modality of hippocampus CA1 region neuron had distinctly changes when compared with that of the controls. Conclusion GLP can improve the activity of SOD, reduce the content of MDA and improve the recessive changes of hippocampus CA1 region neurons. GLP can improve and increase Aβ-injected rats' learning and memory ability.
