摘 要:目的优选从甘草不同器官中提取高质量RNA的方法,为研究甘草药用成分合成酶基因克隆和基因表达奠定基础。方法比较异硫氰酸胍法、柠檬酸钠法所提取的甘草根木质部、根皮部、根茎、叶中RNA的纯度和得率。结果异硫氰酸胍法能从根的木质部、根皮部和根茎中提取出质量较好的RNA,但不能从甘草叶片中提出RNA;柠檬酸钠法能从叶片中提取出质量较高的RNA。结论异硫氰酸胍法和柠檬酸钠法分别是提取甘草根木质部、根皮部和根茎及叶片中RNA的有效方法。Objective To optimize a method for extraction of RNA from different organs of Radix Glycyrrhizae and to provide a base for the study on synthetic enzyme gene clone and gene expression of medicinal components of Radix Glycyrrhizae. Methods The purity and yield were compared of RNA extracted from the xylem, phloem, rhizome and leaf of Radix Glycyrrhizae by methods of guanidine isothiocyanate or sodium citrate respectively. Results RNA could be extracted from the xylem, phloem,rhizome, but not from the leaf of Radix Glycyrrhizae by using guanidine isothiocyanate method, and RNA could be extracted from the leaf of Radix Glycyrrhizae by using sodium citrate method. Conclusion The method of guanidine isothiocyanate or sodium citrate is respectively an effective extractive method for RNA from the xylem, phloem, rhizome, and leaf of Radix Glycyrrhizae.
