摘 要:探讨甾体避孕药对卵巢功能的局部作用,用DES处理未成熟大鼠,取卵巢颗粒细胞(G-C)行体外培养。培养液中分别含有或不含PMSG,FSH,LNG,RU486或ZK98.734以及T。按不同时间结束培养,培养液做激素测定。P和E_2含量测定均采用WHO提供的RIA配对试剂和操作方法。结果:在50mIU/ml PMSG刺激下培养48h,30nM,100nM,300nM的LNG刺激G-C分泌P的产量分别为对照组的224%,220%和251%(P<0.05);100nM,300nM的ZK98.734刺激的P产量分别为对照组的168%和180%(P<0.05)。两者均不刺激大鼠G-C分泌P的基础水平。经48h孵育再加入100nM LNG或ZK98.734培养48h,显示在200nM和1000nM剂量点,LNG使P产量升高5~7倍,ZK98.734使P产量升高3.5倍,RU486未显示类似效应。FSH具有与PMSG的相同刺激效应。结论:LNG和ZK98.734都能促进体外培养的大鼠卵巢G-C分泌P,LNG比ZK98.734的促进作用更强,这种促进作用是通过增强促性腺激素的刺激作用而产生的。The effect of levonorgestrel(LNG) , mifepris-tone (RU486)and lilopristone (ZK98. 734) was studied on granulosa cells in vitro. Immature female rats were treated with diethylstilbestrol (DES)for 3 days. Granulosa cells were collected from ovary and cultured in 0. 5ml McCoy' s medium, with or without pregnant mare' s serum go-nadotropin (PMSG) . follicle stimulating hormone (FSH), testosterone (T ), LNG, RU486, or ZK98. 734. The media were collected after 48h incubation. In some experiments .after granulosa cells incubated with PMSG(1. 6,8,40,200,1000nM)for 48h,100nM LNG or ZK98. 734 were added and continuously incubated for another 48h. Progesterone (P) and estradiol(E2) concentrations in the media were measured by RIA.The results showed that: (1)With the treatment of 50mIU/ml PMSG, low doses LNG or ZK98. 734 (3, 10nM) showed no response, but in high doses of LNG ( 30, 100, 300 nM ) a 224%, 220%and 251% increase in P production resulted respectively (P<0. 05) (100, 300nM of ZK98. 734 resulted in a 168% and 180% increase(P<0. 05). All doses of RU486 had no effect on P secretion with or without PMSG treatment;(2)In double 48h incubation experiments, the results showed that granulosa cells incubated with PMSG at doses of 200,100 nM/ml, produced P upto 5 - 7 times by LNG (the increase was 3. 5 times by ZK98. 734. FSH had similar stimulation as PMSG; (3) When FSH and T were added at the same time .different doses of LNG(2- 100 nM)all promoted P production upto more than 30nmol/L levels,but no effect on E2 le vels.We concluded that LNG and ZK 98. 734,but not RU486,can increase the P production by rat granulosa cells cultured with PMSG or FSH. LNG had stronger action than ZK98. 734. This increase in P production was acting through the enhance- ment of gonadotropin stimulation.
