摘  要:目的:观察熟地黄的抗焦虑作用,并初步探讨其机制。方法:实验于2003-09/2005-09在河南中医学院动物实验中心和中药研究室进行。60只昆明种小鼠,按体质量随机分为5组:对照组,地西泮组,熟地黄高、中、低剂量组各10只。对照组每天灌胃生理盐水;地西泮组前4d每天灌胃生理盐水,后3d灌胃地西泮2.5mg/(kg·d);熟地黄高、中、低剂量组灌胃熟地黄分别为12,8,4g/(kg·d),连续7d。7d后开始进行小鼠明暗穿箱实验、小鼠高架十字迷宫实验,观察记录10min内小鼠穿箱次数,分别记录实验期5min内小鼠进入开放臂和封闭臂的次数、停留时间及在两臂滞留的总时间。计算小鼠进入开臂的次数和在开臂滞留时间分别占总入臂次数和总入臂时间的百分率,以此作为评价药物抗焦虑作用的指标。取对照组、地西泮组及熟地黄中剂量组小鼠行为学观察后立即处死,剥离大脑,用薄层扫描法测定γ-氨基丁酸、谷氨酸含量;用免疫组化法测定GABAAR1、N-甲基-D-天冬氨酸受体1表达。组间比较采用t检验。结果:60只小鼠均进入结果分析。①与对照组比较,地西泮、熟地黄中剂量组能显著增加小鼠的穿箱次数[(17.1±4.6),(32.5±13.7),(26.7±4.1)次,P<0.01],地西泮与熟地黄中剂量组比较,差异不显著。②与对照组比较,地西泮组进入开臂的时间百分率、次数百分率显著增加[(4.51±2.31),(10.83±6.92)%;(14.46±10.71),(25.80±3.61)%,P<0.05,P<0.01],熟地黄中剂量组进入开臂的次数百分率显著增加[(14.46±10.71),(25.42±6.86)%,P<0.01]。地西泮组与熟地黄中剂量组比较,差异不显著。③与对照组比较,熟地黄中剂量组、地西泮组小鼠脑组织中谷氨酸含量显著降低[(571.70±52.27),(477.41±41.10),(454.64±32.60)μg/g,P<0.01],γ-氨基丁酸含量显著提高[(182.52±8.65),(239.56±18.00),(295.10±14.83)μg/g,P<0.01]。熟地黄中剂量组与地西泮组比较,差异不显著。④与对照组比较,AIM: To study the anti-anxiety effects of prepared rehmannia root and investigate its mechanism. METHODS: The experiment was conducted in the Animal Experimental Center and Traditional Chinese Medicine Laboratory of Henan College of Traditional Chinese Medicine from September 2003 to September 2005, Sixty Kunming mice were randomly divided into 5 groups by body mass: normal control group, Diazepam group, high, middle and low doses prepared rehmannia root groups with 10 mice in each group, The control group was intragastric infused with normal saline everyday; the Diazepam group was infused saline in the first 4 days following by Diazepam 2.5 mg/kg per day for 3 days; the high, middle and low doses prepared rehmannia root groups were infused with 12, 8, 4 g/kg per day prepared rehmannia root for 7 days. Then the elevated plus-maze test and light-dark transitions test were performed to observe the times of mice passing the box in 10 minutes, and record the times of the mice entering the open arms and close arms, stay time and total time in two arms in 5 minutes. The percentage of the times entering the open arms and stay time in total times and time in arms were calculated and served as the index to evaluate the anti-anxiety effect. The mice from the control, Diazepam and middle dose groups were killed after behavioral observation and the brain was removed to measure the content of gama-aminobutyfic acid (GABA) and glumatic acid (Glu) in the brain by thin-layer chromatography, and observe the expression of GABAAR1, N-methyl-D-aspartate receptor 1 (NMDAR1) by immunohistochemical means. Comparison between groups was performed by t-test. RESULTS: All the 60 mice were involved in the result analysis. ①Compared with the control group, the middle dose of prepared rehmannia root and Diazepam could distinctively increase the passing times in the lightdark transitions test [(17.1±4.6), (32.5±13.7), (26.7±4.1) times, P 〈 0.01].
