摘  要:目的研究红光激光对血管内皮细胞一氧化氮(NO)分泌及表达的影响。方法应用红光激光分别照射体外培养的血管内皮细胞10 min、20 min和30 min后,分别于照射后15 min、30 min、1 h、3 h和6 h检测培养上清液中NO的含量,并应用免疫组化方法检测内皮细胞内诱生型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)的表达,结果2 mW红光激光分别照射10 min、20 min、30 min可使内皮细胞内eNOS表达增强,分泌NO水平上升,并于1 h后达峰值,随后逐渐降低,与对照组比较差异有统计学意义(P< 0.05),其中照射30 min组作用最明显;红光激光照射对内皮细胞内iNOS的表达各组比较差异无统计学意义(P>0.05)。结论红光激光可通过增强内皮细胞内eNOS的表达来促进NO的分泌;红光激光对内皮细胞内iNOS的表达无明显影响。Objective To observe endothelial cells secreting and expressing nitric oxide by using red laser irradiation (RLI) on cultured endothelial cells. Methods Cultured endothelial cells were irradiated with a red laser for 10, 20, and 30 minutes, respectively, and the concentration of nitric oxide in the cell supernatant was measured after 15 min,30 min,1 h,3 h and 6 h. In addition, the expression of inducible nitric oxide synthetase (iNOS) and endothelial nitric oxide synthetase (eNOS) was measured through immunohistochemical staining. Results Compared with that in the control group, the expression of eNOS by the endothelial cells and the concentration of NO were stimulated by 2 mW RLI for 10 min, 20 min and 30 min, and reached a peak at 1 h, then declined gradually. The expression of iNOS, however, showed no significant difference. Conclusion RLI can increase NO concentrations in endothelial cells by stimulating the expression of eNOS rather than iNOS.
