摘 要:目的探讨p38丝裂原活化蛋白激酶(p38MAPK)在低氧引起的鼠肺动脉平滑肌细胞增殖中的作用。方法采用细胞培养方法,免疫印迹技术,流式细胞仪方法测定在低氧情况下p38 MAPK的表达及其细胞周期的变化。结果低氧情况下细胞中p38 MAPK表达增加,细胞呈现增殖性变化。结论p38 MAPK在低氧情况下参与了鼠肺动脉平滑肌细胞增殖,具有重要的病理生理学意义。Objective To study the expression of p38 mitogen activated protein kinase (p38 MAPK ) in rat pulmonary artery smooth muscle cells (PASMCs) under hypoxia, Method Cultured rat HPASMCs were divided into two groups ① control group; ②48 hours hypoxia group. Expression of ( p38 MAPK ) proteins were detected by Western blot and cell cycle was analyzed by flow cytometry. Result Western blot showed that p38 MAPK protein expression under hypoxia increased and the analysis of cell cycle by flow cytometry showed that the proportion of cells at G1 phase decreased significantly while the cells at S phase increased significantly in hypoxia group ( p 〈0. 01 ). Condusions The results suggested that hypoxia promoted p38 MAPK protein expression in HPASMCs, which played an important role in pulmonary vessel structural remodeling in PASMCs proliferation.
