摘 要:目的对钒酸氧化法、二氯苯胺法和改良J-G法测定血清胆红素进行方法学评价,探讨溶血、脂血对钒酸氧化法测定血清胆红素的干扰影响。方法用钒酸氧化法测定血清总胆红素,按NCCLS评价方案系统评价其重复性、回收率、线性范围和溶血、脂血对血清胆红素测定的干扰影响,并与二氯苯胺法和改良J-G法测定血清总胆红素结果进行比较。结果钒酸氧化法与二氯苯胺法和改良J-G法有良好的相关性(Y1=1.06X-0.32,r=0.94;Y2=0.93X+3.26,r=0.98);钒酸氧化法测定总胆红素的线性可达684μmol/L,批内变异为1.25%,日间变异为3.14%,回收率为102.5%;血红蛋白和甘油三酯浓度分别达6.0g/L和8.0mmol/L,无明显干扰。结论钒酸氧化法测定血清胆红素不仅简单、快速、稳定,而且线性范围宽,能抵抗溶血、脂血干扰,值得推广应用。Objective To compare vanadate oxidation with DAC and modified diazutate J G methods in determination of bilirubin and to investigate interference ability of haemolysis and lipaemia. Methods The serum total bilirubin was measured by vanadate oxidation, DAC and modified diazotate J-G methods. The repetitive experiments, recovery rate, linear range and interference ability were assessed according to the standard method of NCCLS, and relativity between vanadatc oxidalion with DAC and modified diazotate JC- were calculated. Results There was a strong concordance between vanadate oxidation with DAC as well as modified diazotate J-G methods, Y1 = 1.06X-0. 32,r=0. 94 Y2 =0.93X + 3.26, r= 0. 98, respectively. The total bilirubin measured using vanadate oxidation were 684μmol/L for linear range, 102.5% for recovery rate, 1.25% for intra-batch CV and 3.14% for out-date CV, respectively. There were no significant interference when hemoglobin less than 6.0 g/L and lipidemia less than 8.0 mmol/L. Conclusion The method of van,,dine oxidation is single, rapid, and stable. It has a wider linear range and an ability to interference of hemoglobin and chylomicronemia. There is an important value of clinical application.
