摘 要:目的:研究重组人白细胞介素6(rhIL-6)对原代培养大鼠肝细胞分泌纤维蛋白原(Fibrinogen,Fgn)的影响及蛋白激酶C(PKC)抑制剂Ro31-8220对枯否细胞分泌IL-6的作用。方法:以酶联免疫吸附测定法(ELISA)测定Fgn;测定I-6采用[3H]-TdR掺入增殖法。结果:大鼠肝细胞(3×1O6/ml)与rhIL-6(0~100kU/L)孵育24h,刺激肝细胞分泌Fgn增加;Ro31-8220(0.1~10μmol/L)显著抑制枯否细胞释放IL-6。结论:提示IL-6是调节纤维蛋白原合成的重要细胞因子,推测PKC抑制剂可通过抑制枯否细胞分泌IL-6来降低Fgn的分泌。To investigate the effect of interleukin-6 (IL-6) on the fibrinogen production from rat primary hepatocytes culture and Ro31-8220 on the IL-6 production from Kupffer cells. Methods: Enzyme link immunosorbent assay(ELISA) was used to determine fibrinogen levels in hepatocyte culture and IL-6 level was determined by [3H]-TdR incorporation assay. Results: IL-6 (0~100 kU/L) dose-dependently enhanced fibrinogen production after cultured with hepatocytes(3×106/ml) for 24 h and Ro31-8220 (0.1~10 umol/L) markedly reduced IL-6 secretion. Conclusion: IL-6 plays a crucial role in modulation of fibrinogen and PKC inhibitors could decrease fibrinogen production through the inhibition of IL-6 secretion from K upffer cells.
