胃癌围手术期血清IL-10、IL-12浓度的变化及其对CIK细胞的影响

摘　　要：目的检测胃癌患者围手术期血清白细胞介素(IL)-10、IL-12的浓度变化及其对细胞因子诱导的杀伤细胞(CIK细胞)表型和活性的影响,并探讨其意义。方法用ELISA法检测22例胃癌患者(胃癌组)围手术期血清IL-10、IL-12的浓度;用MTT法测定其围手术期血清对CIK细胞活性的影响;用流式细胞术对其围手术期血清培养的CIK细胞表型(CD3、CD56分子)进行分析,以16例健康人作为对照组。结果胃癌组患者血清IL-10浓度升高,术后血清IL-10浓度逐渐下降,但仍高于对照组;血清IL-12浓度降低,术后血清IL-12浓度逐渐升高,但仍低于对照组;其血清培养的CIK细胞活性及CD3+CD5+6CIK细胞表型平均百分率术后高于术前,均低于对照组。结论胃癌细胞可分泌IL-10,抑制IL-12,进而对CIK细胞表型的表达产生影响,活性产生抑制。术后血清IL-10浓度逐渐下降,IL-12浓度逐渐上升,CIK细胞活性逐渐增高。Objective To study the changes of serum interleukin（IL） - 10 and IL - 12 levels in patients with gastric cancer within perioperative period, and evaluate the influence of them upon the activity and phenotype of cytokine induced killer （CIK） cells. Methods The levels of serum IL - 10 and IL - 12 in 22 patients of gastric cancer were examined by ELISA assay preoperatively. The activity of CIK cells were measured by MTY assay and 16 healthy people as health group. The phenotype （ CD3, CD56 ） of these CIK cells were analysed by flow cytometry. Results In patients with gastric cancer, the serum IL - 10 levels were decreased significantly in three periods respectively, but they were all higher than that in health group. Meanwhile, the serum IL - 12 levels were increased significantly in the correspond three periods respectively, but they were all lower than that of health group. CIK cells were cultured by the correspond three periods serum of the patients, the activity of CIK ceils and the percentage of phenotype （ CD3 , CD56 double positive cell） of CIK cells were higher than those in preoperation, which were all lower significantly than that of health group. Condusion Gastric cancer cells can secrete IL - 10, and repress IL - 12, thus affecting to the CIK cells. Their immune function will be recovered with getting rid of immunosuppressive factors after tumor eradicated curatively, so the levels of IL - 10 and IL - 12 and the activity of CIK cells will be return to normal level gradually.
