摘 要:[目的]观察糖肾汤对氧化低密度脂蛋白(ox-LDL)诱导的大鼠系膜细胞增殖及TGFβ1mRNA表达的作用。[方法]培养大鼠系膜细胞,分为空白对照组、ox-LDL组、糖肾汤高、中、低剂量组和罗格列酮组,培养24h后,采用MTT法检测细胞增殖,用RT-PCR技术检测系膜细胞TGFβ1mRNA表达情况。[结果]50μg/ml的ox-LDL可促进细胞增殖和TGFβ1mRNA表达;糖肾汤和罗格列酮具有明显抑制ox-LDL诱导的MC增殖和TGFβ1mRNA表达增加的作用。[结论]糖肾汤可抑制ox-LDL诱导的MC增殖和TGFβ1表达的增加,从而在糖尿病肾病的治疗中起作用。[Objective] To investigate the effect of Tangshentang on the proliferation and expression of TGFβ1 mRNA of cultured rat kidney masengial cell (MC) induced by ox LDL. [Method] The cultured kidney MC were divided into control group, ox LDL group, Tangshentang group (respectively high, medium and low group)and Rosiglitazone group. The mRNA expression was tested by RT-PCR. The proliferation of MC was tested by MTT. [Result] Ox-LDL can increase the cell proliferation and the expres sion of TGFβ1 mRNA. Tangshengang and Roaiglitazone significantly inhibited the enhanced levels of TGFβ1 expression and the proliferation of rat kidney MC stimulated by ox LDI.(50μg/ml). [Conclusion] Tangshengang plays a role in the treatment of DN through reducing the levels of TGFβ1 mRNA expression and cell proliferation induced by ox-LDL.
