摘  要:报道了以氨丙基硅胶为载体,将其表面醛基化后通过亚胺键共价键合牛血清白蛋白,再用两种硅烷化试剂氨丙基三乙氧基硅烷和正辛基三甲氧基硅烷进行聚合,合成表面印迹牛血清白蛋白分子印迹聚合物的方法。本方法以共价键印迹蛋白质,用草酸洗脱。表面印迹有利于大分子蛋白质向印迹位点的扩散和再结合,合成的印迹聚合物对牛血清白蛋白的吸附率达44.5%,而其它蛋白的吸附率小于17%,显示对模板蛋白具有特异吸附能力。Molecular imprinting of bovine serum albumin (BSA) on silica surface is described using a covalently bonded template method. For polymerization, two kinds of organic silane (3-aminopropyltrimethoxysilane: APS and n-octyltrimethoxysilane: OMS) were used. Aminopropylsilanized silica gel was first aldehyde functionalized with glutaraldehyde and then reacted with BSA by forming imine bonds to covalently immobilize the template protein. Polymerization occurred for siloxane-silica in the presence of the template protein. After removal of the template with oxalic acid, the BSA imprinted polymer on silica surface was obtained. Adsorp- tion experimental results indicate that the surface imprinted polymer show high selective binding for the template BSA over the other tested proteins. One of the main advantages of the method is its binding sites located on or close to the surface and thus having good site accessibility towards the target protein.
