摘 要:为探讨不同培养基及共培养对克服小鼠早期胚胎体外发育阻滞的影响,用不同成分组成的5种常用培养液对小鼠早期胚胎进行体外培养,后以KSOM作为基础培养基,KSOMFBS为对照,分别与小鼠输卵管上皮细胞(MOEC)和卵丘细胞(CC)进行共培养,观察从2细胞发育到4细胞的胚胎数和囊胚数的变化。结果发现5种培养液中从2-细胞发育到4-细胞的胚胎率为30.5%~43.5%,差异不显著。KSOM组囊胚率为40.9%,显著高于其他各组(27.9%~30.0%)。共培养后,各组中克服发育阻滞的胚胎数和囊胚数均有显著提高,其中添加胎牛血清组4细胞胚胎数和囊胚数显著高于不添加组,输卵管上皮细胞组显著高于卵丘细胞组。结果显示共培养对克服小鼠2-细胞发育阻滞作用明显,添加胎牛血清效果明显好于不添加,输卵管上皮细胞共培养效果好于卵丘细胞。This paper describes the influence of five different culture mediums and co-culture approaches on mouse preimplantation embryos to overcome developmental block in vitro. Four different common mediums were used to culture mouse preimplantation embryos in vitro, and then select KSOM as basal medium, KSOMFBS control medium, separately combined with mouse oviduct epithelial cells (MOEC) or cumulus cells (CC) to co-culture the embryos. The changes of embryo numbers from 2-cell to 4-cell and blastocysts numbers were observed. The result showed that embryo development rate from 2-cell to 4-cell ranged from 30.5%-43.5% and the difference was not remarkable. The blastocysts rate in KSOM group was 40.9%, higher than other groups (27.9%-30.0%). After coulture, the numbers of 4 cell embryos and blastocysts were remarkably raised in each group and the numbers of 4-cell embryos and blastocysts in the group added fetal bovine serum(FBS) were significantly higher than no serum group, the group with oviduct epithelial cells higher than cumulus cell group. The results suggested co-culture has obvious effect for embryos to overcome in vitro development block. To add FBS has better effect than no FBS and MOEC co-culture better than CC.
