摘 要:探讨了妊娠早期山羊胎儿IL-4水平与外周血单核细胞(PBMC)增殖活性的关系。用MTT法,检测孕早期山羊胎儿匀浆上清液(FHS)在不同浓度下对PBMC转化增殖活性的影响,设立孕130d胎儿血清(FS)及FHS+PHA-P对照组;检测蛋白质浓度在1×10^-4mg/mL时IL-4的含苗,并与转化后的含量进行比较。结果:妊娠早期,FHS对PBMC转化活性的影响随着妊娠的发展而逐渐降低,但均低于妊娠后期对照组。FHS蛋白质浓度在1×10^-4~1×10^-5mg/mL时,对PBMC的转化增殖效果均较好(P〈0.05),且与PHA—P具有协同作用;妊娠早期FHS的IL-4水平随着妊娠的发展呈现上升的趋势,且均高于FS对照组,FHS的IL-4水平与转化后无显著差异(P〉0.05)。妊娠早期FHS IL-4水平的上升与PBMC转化活性下降之间呈负相关关系,正常妊娠早期主要是Th2型细胞因子在起作用,因此可以说明,PBMC转化活性的下降可能是IL-4发挥免疫抑制作用的结果,进而说明,妊娠早期胎儿可能通过自身分泌一些Th2型细胞因子,在母体子宫局部免疫中发挥免疫抑制作用,使其自身不被母体排斥,使妊娠能够顺利进行。To study the effect of IL-4 level of Goat fetus during early pregnancy on Proliferation of Periferal Blood Mononuclearcells (PBMC). Detecting the Fetal Homogenate Supernatant (FHS)' s effect on Proliferation of PBMC during early pregnancy at different protein concentrations by MTT essay, with Fetal Serum(FS) and FHS cooperated with PHA-P as control group; Detecting the IL-4 level of FHS when protein concentration is 1×10^-1 mg/ mL, compared with it after transformation. The effect of FHS on lymphocyte proliferation cut down and the IL-4 the concentration of FHS is 1×10^-4 mg/mL the pregnancy goes on level of FHS is strengthen when during early pregnancy, but it is contrary in the later stage of pregnancy; Lymphocytes remarkable proliferated when protein concentration is 1×10^-4 -1×10^-5mg/ mL(P〈0.05), and transformation effect of FHS cooperated with PHA-P was much better than it is single(P〈0.05). It has no difference between IL-4 level of FHS and it after transformation(P〉0.05). Conclusion. The IL4 level of FHS is strengthen and it is contrary with the effect of transformation as the pregnancy goes on during early pregnancy, because of Th2 cytokines play the dominated role during early pregnancy, it is indicated that the cutting down of PBMC's transformation effect is prosibly caused by IL-4 that plays the immunosuppressive role during early pregnancy. Furthermore, it is indicated that Fetus plays the immunosuppressive role in uterine local immunity during early pregnancy through secreting some Th2 cytokines to avoid the immunological rejection by parent, and that makes the pregnancy to proceed smoothly.
